"Noun
One who is excessively concerned with being or appearing to be proper, modest, or righteous."

Its like a goody goody. They won't do anything wrong. Or for example, won't kiss anyone or go out w/ anyone.

It means she wont do anything sexual.
Like say he wanted to makeout but she never has and doesnt want to, that would generally be known as prude.

A prude is a person who is overly modest or prim and proper. It is generally not a positive thing to say to a person.
It depends on why he called her a prude. Does he want her to do something she is not comfortable doing, or is she lacking a sense of humour, or unable to let go in a social situation and enjoy herself?
Hope that helps.
