I'm assuming you're dealing with literature homework.

The plot of a story is the sequence of events (rising actions, climax, falling actions, etc.), and the main storyline. What it's about.

The theme of a story is the "moral" or "lesson" learned from the story.

Xineph answered Sunday January 21 2007, 1:37 pm:
A plot is a storyline, the collected stream of events that happens through the story. Example: The plot of "The Three Little Pigs" is about a big bad wolf who goes around blowing houses down....

Themes are ideas, concepts, and undertones that carry through the plot. Example: Themes of "The Three Little Pigs" include hard work, being prepared, and helping others in need.

Seriously though, you could have looked all these up on dictionary.com. Advicenators is supposed to be your last resort.
