For the most accurate reading you take them every single day of your menstrual cycle except when you're having your period. It's much like a pregnancy test in that you'll just put a little urine on the strip and wait so many minutes for the results. The ones I took showed a purple line if I was ovulating and no line at all if I wasn't.
For the best results to figure out when you usually ovulate you should take these tests for 6 months straight.
You can find the over-the-counter tests in a variety of "sizes" in that some will be 7 tests online (for those women who are nearing the end of the 6 month term, usually) or up to 20 tests. Go with the one with the most in it so you can test every day. After the first few months you should be able to dwindle the day you ovulate down to about a weeks time and then you can start purchasing the boxes with less tests, if you get the idea.
So, no testing when you're on your period.
