If your going to do a speech I think the most effective one is to bring up a couple good times you have shared with her and how great having her in your life has been. I also think you should mention the groom, maybe describe the day you met him and what you thought. It's always good at the end to say something along the lines of like this day came so fast I can't believe she's already getting married. Or like. "I'm happy to gain a new member to the family and glad that my sister has finally found that special someone." Also, wish them happiness and long lasting love, something sweet and meaningful. I'm sure if you put your heart into it you'll come up with a great idea that everyone will love!

Hope I helped!
:)
