I dont know how to talk my best guy friend about "us." How do i talk to him, call him to meet somewhere?
Should I email him, i am shy around him even though he wants to hook up with me.

Ask him to meet you somewhere where you two can be alone. Somewhere quiet, not a movie. More like the park or a restaurant. Tell him exactly how you feel. If he feels the same way, he will tell you. If he doesnt he will also tell you, but he will say that you two are very close friends. Dont take this answer seriously because he may just not want to ruin your relationship.
